Ipamorelin’s endocrine measurements
The endocrine result is elegant. Its particular measurements matter even more once the bowel and sensitivity experiments join the picture.
Rat cells, rats and swine
The founding characterization of ipamorelin measured growth-hormone release in primary rat pituitary cells, anaesthetised rats and conscious swine. Ipamorelin released growth hormone potently while ACTH and cortisol did not rise above the level seen with growth-hormone-releasing hormone, even at doses more than 200 times the GH ED50. This was an acute characterization.
The specificity of that achievement delights me. The investigators wanted a growth-hormone response with less stimulation of the other hormone measurements in that comparison, and the experiment supplied one. The phrase selective growth-hormone release has an actual experimental subject.
The rat sensitivity paper asks about ghrelin-receptor involvement in behavioural responses. These results can coexist comfortably. Hormonal selectivity in an acute comparison leaves a whole body available for other investigations.
Intravenous ipamorelin in men
In the human pharmacokinetic and pharmacodynamic study, healthy male volunteers received intravenous infusions. The model described a single discrete growth-hormone pulse peaking at about 40 min, with an approximately 2 h terminal half-life for ipamorelin.
A pulse and a half-life describe different things: the timing of the hormone response and the disappearance of the administered compound from plasma. Those timings are satisfying in their own right; the gut papers draw my curiosity toward symptoms and function. Meal tolerance after surgery had to face its own randomized trial.
